Never done it. I do have day care but never weekend care of just for a few hours. My sister's friend need care for only 4 hours for her 2 kids ( 10 & 4) how much should I charge? Thanks!

Depends on area. My daughter would charge $8/hr per child. -

Regularly, I'd consider it part of daycare and I'd charge my daily rate plus a charge for weekend.
Once in a while, I'd charge $12/hr.
And probably, unless it was an emergency or I wanted to make some $, I'd pass it off on to an actual 'babysitter'.
